After first installation, WinHex puts out a warning that a open file has been modified externally. As I wanted WinHex to always automatically refresh the file I checked the 'Do not warn me anymore' box. Now it doesn't warn me anymore and also doesn't auto-refresh.
How can I undo this, or even better make WinHex auto-refresh?

You can reset all settings with Help | Setup | Initialize, but cannot make WinHex auto-refresh without prompting.
